Effect of Cardioselective Beta Blocker Metoprolol on EpileptiformActivity Induced by Penicillin in Rats

Abstract

AIM: Limited information is available on the effects of thecardioselective beta-blocker metoprolol, which is commonly usedin the treatment of cardiac and hypertensive problems in epilepsypatients, on epileptic activity. The aim of this study is to shed lighton the clinical review of metoprolol treatment in patients withepilepsy by determining whether its effects on epileptic activity areproconvulsant or anticonvulsant.

METHODS: For this purpose, fourteen male Sprague-Dawley ratswere divided into two groups with 7 rats in each group. The firstgroup (SHAM group) received a single dose of penicillin (500 IU, 2.5μL, i.c.) and 0.9% saline (0.5 mL, i.p.). The second group (Treatmentgroup) received a single dose of penicillin (500 IU, 2.5 μL, i.c.) andmetoprolol (0.5 mL, 50 mg/kg, i.p.). Metoprolol was administered30 minutes after the penicillin injection, as the epileptic spikeswere expected to become prominent. Each rat was fixed in astereotaxic apparatus and ECoG recordings were taken for 240minutes with electrodes placed on the brain tissue. The evaluationof the recording data was based on the frequency and amplitudesof the epileptic spikes. The average frequency was calculated asspikes/min, and the average amplitude was calculated as μV in 10-minute periods and included in the statistics.

RESULTS: The two independent groups, SHAM and Treatment,were statistically compared based on the average spike frequencyand amplitude values in 10-minute periods, and no statisticallysignificant result was obtained.

CONCLUSIONS: In conclusion, further studies focusing on differentdose ranges and cellular mechanisms in the brain are needed tofully understand the effect of metoprolol on epileptic activity.
