Özet

Aim: Stachys subnuda (SS) is a species belonging to the Stachys genus within the Lamiaceae family. It is known that species of the Stachys genus are rich in flavonoids, and their antioxidant properties have been demonstrated in previous studies. Additionally, some species have shown cytotoxic and apoptotic effects on breast cancer cell lines. However, studies on the SS species are quite limited. SS is endemic to Turkey and is commonly known as "mountain tea" among the local population. Traditionally, SS has been used for skin, stomach, respiratory, and kidney ailments.

Methods: The aerial parts of the SS plant were collected from Tunceli. The methanol extract of SS (SSM) and its hexane (SSH), chloroform (SSC), ethyl acetate (SSEA), and aqueous methanol (SSAM) fractions were prepared at the Faculty of Pharmacy, Marmara University. The extracts were evaluated for cytotoxicity and apoptosis with CCK-8 and JC-1 mitochondrial membrane potential tests. GraphPad Prism (version 8.0.1) was used for statistical analysis. One- way ANOVA followed by Tukey's test was performed.

Results: When SSM, SSH, SSC, SSEA, and SSAM extracts were applied to MCF-7 breast cancer cells at various concentrations (0, 10, 25, 50, 75, 100 µg/mL) for 24 hours, the IC50

values were calculated as 30.45, 16.44, 77.73, 70.07, and 34.18 µg/mL, respectively. The selected SSH extract statistically significantly depolarized the mitochondrial membrane potential (p < 0.05).

Conclusions: The cytotoxic and apoptotic effects of the SS species on the MCF-7 cell line were investigated for the first time, suggesting that SSH may have therapeutic potential.
